Molecular Mechanisms of Malaria Parasite Invasion
Objectives
The overarching aim of this research is to understand the biological interface between malaria parasites and their human host. This research program will characterize the role of PvRBPs in P. vivax invasion, determining the structure-function requirements for PvRBPs in red blood cell recognition and characterize new host-pathogen interactions that allow P. vivax to enter young red blood cells.
A detailed understanding of PvRBPs’ molecular and structural function will progress the development of new inhibitors and monoclonal antibodies that block P. vivax invasion and reveal their role during the dynamic process of parasite invasion.

Malaria parasites are obligate intracellular parasites exquisitely adapted for invasion and survival within red blood cells. Parasite entry into red blood cells involves specific interactions between parasite adhesins and their cognate red blood cell receptors. For the two most common human malarial parasites Plasmodium falciparum and Plasmodium vivax, there are two orthologous families of parasite adhesins that govern entry into red blood cells; Reticulocyte-Binding Homolog in P. falciparum (PfRh) and Reticulocyte-Binding Proteins in P. vivax (PvRBPs). While these protein families have been increasingly implicated in mediating cell and host tropism, the molecular mechanisms of specificity and their respective host-pathogen interactions are not very well understood.
