Harnessing a potent multi-affinity antibody platform to inhibit malaria infection

Malaria remains a global health concern, and is responsible for approximately 400 000 deaths annually, primarily in children under 5. Malaria is transmitted through the skin by the bite of an infected mosquito. Widely-used methods of control, including bed nets, insecticides and drug treatments have failed to eliminate malaria, and increasing resistance to drugs and insecticides highlights the need for new tools for prevention. Traditional antibody therapies – the delivery of an antibody into the bloodstream – are a promising option, but are limited by the need for high doses that do not persist to provide long-term protection. To overcome these barriers, a molecule has been developed that combines multiple antibodies to form an extremely potent drug that will allow the delivery of lower, cost-effective doses to prevent malaria infection. Finally, the molecules will be modified to concentrate drug at the site of malaria transmission in the skin to prevent infection. The goals align with Canada’s humanitarian efforts and contributions to infectious disease eradication, including through the Global Fund to fight AIDS, Tuberculosis and Malaria. These diseases disproportionately harm women and girls, and effective preventive drugs will contribute to Canada’s ongoing commitment to gender equality. It has already been shown that this method dramatically increases the potency of antibodies against HIV and SARS-CoV-2. The format is easily modified, and its further development has the potential to contribute to the fight against emerging infectious diseases and future pandemics, allowing rapid responses to global health threats.
