Evaluation of the phenotypic profile of chloroquine metabolization in a city endemic for P. vivax malaria in the Brazilian amazon region

This study will aim to evaluate the molecular characterization of the CYP2C8 gene to identify genotypic and phenotypic profiles of chloroquine metabolism in 150 DNA samples obtained from patients living in the municipality of Cruzeiro do Sul, in the state of Acre.
Malaria is one of the main endemic diseases in Brazil, affecting the quality of life of populations in tropical and subtropical regions, even though it is preventable and treatable. In this context, although several studies associate therapeutic failures of the combination of Chloroquine and Primaquine with polymorphisms of the genes that metabolize antimalarials (CYP2C8 and CYP2D6), it is necessary to deepen the knowledge about the genotypic and phenotypic profiles of these metabolizers and the possible association of these polymorphisms with recurrence of malaria by P. vivax in Brazil, especially in the Amazon region. The data generated by this study will allow the analysis of the population and haplotype allele frequency (individual and population), and define the phenotypic prediction for the metabolization of Chloroquine for each participant and the studied population.
