This project aims to investigate the heterochromatin control mechanism in the malaria parasite at the molecular level.
Unlike viruses and bacteria, Plasmodium malaria is difficult to establish a treatment method because it avoids host immunity by controlling gene expression with heterochromatin peculiar to eukaryotes, which is a major obstacle to malaria control. Interestingly, malaria protozoa perform epigenome regulation by heterochromatinization similar to the host immunoavoidance mechanism in sexual differentiation, which is the conversion from asexual to sexual reproduction. Therefore, the applicant focused on the expression control of the sex differentiation factor AP2-G, and used the proprietary technology of genome editing and transcriptional control evaluation system using artificial chromosomes to investigate the heterochromatin control mechanism in Plasmodium at the molecular level.
