Does the cytotoxic phenotype of Th cells during malaria infection mediate pathogenesis in an infected host?
Objectives
This project will investigate the role of T-cells in the generation of severe disease by using CHMI-derived samples to determine if T-helper cells are capable of mediating damage to the host – either directly or via an intermediate signal provided to another domain of the immune system.
Malaria infection doesn’t always result in severe disease, but a trend exists between severity and infection history. The majority of severe cases emerge in individuals with no or limited prior exposure. Risk of severe disease decreases with repeated rounds of infection. Using a controlled human malaria infection (CHMI), internal data from the host lab identified that T-helper cells present a cytotoxic phenotype during primary infection. The results of these experiments will be compared between primary and secondary/tertiary infections, to investigate the change in T-cell phenotype with repeated infection. Finally, making use of samples derived from individuals naturally infected with malaria during a trip to an endemic region, this project will attempt to produce a model for predicting disease severity from T-cell phenotype.
