Rationale and Abstract

In order to clarify changes in clinical resistance to artemisinin over time in Kinshasa, Democratic Republic of the Congo, a survey of clinical artemisinin-resistant malaria was conducted by monitoring the parasite infection rate before and after administration of artemisinin in patients with falciparum malaria in three districts of Kinshasa. Before administering the drug, blood is collected and the malaria parasites contained in the blood are frozen and preserved. If there is a case showing clinical resistance to artemisinin, a culture strain will be established. In addition, in order to clarify the distribution of artemisinin clinical resistance, similar artemisinin clinical resistance evaluations will be conducted in the northeastern part of the Democratic Republic of the Congo, and arrangements will be made for blood collection, transportation, and cryopreservation to establish malaria parasite cultures. Plasmodium falciparum culture strains will be established from previously obtained blood samples from cases that appear to be clinically resistant to artemisinin, and in vitro susceptibility tests to various antimalarial drugs, including artemisinin, will be conducted and the entire genome sequence will be determined.
