Development of an innovative direct protein introduction method into living cells using membrane permeation promotion technology

Objectives

This project aims at the development of an innovative direct protein introduction method into living cells using membrane permeation promotion technology.

Principal Institution

Ehime University, Japan

Principal Investigators / Focal Persons

Eizo Takashima

Rationale and Abstract

The system for introducing genes into cells has already been established, but the “system for introducing proteins into cells” is still under development. Therefore, by combining the cell membrane permeation tag [Technical seeds 1] derived from Plasmodium malaria protein and the membrane permeation promoter [Technical seeds 2] derived from the theory of membrane curvature, a large protein of about 50 kDa, which was impossible until now, can be realized. This will establish a system that can be directly introduced with high efficiency without damaging nuclear cells. A wide range of biotechnology fields, such as intracellular protein structure and function analysis using modified proteins, intracellular molecular localization / function inhibition analysis using antibodies, and protein amount-dependent reaction analysis by changing the amount introduced. And this study will lay the foundation for membrane permeation experimental kits available across.
