Do genetic polymorphisms in the blood group Glycophorin affect malaria erythrocyte invasion?
Objectives
This study will attempt two approaches to answer the question of how polymorphisms (blood types) in the GYP molecule group affect erythrocyte invasion of malaria parasites.
- Analysis of the correlation between malaria severity and blood type in populations in endemic areas.
- Observation of the efficiency of invasion of cultured malaria parasites into cells (erythrocytes) transfected with each allele of the gene encoding the GYP molecule.
The purpose of the above analysis is to clarify the significance of the GYP polymorphism in malaria aggravation, and to contribute to the development of a preventive treatment for severe malaria, which is directly linked to malaria death.
Glycophorin (GYP) molecules are expressed on the surface of erythrocytes and determine classical blood types such as MN, Ss, Gerbich, and Dantu. Recently, it was reported that GYP molecules function as receptors involved in erythrocyte invasion of malaria parasites. In 2021, stored DNA extracted from 50 severe malaria patients was first targeted, 100 asymptomatic malaria patients, 100 febrile malaria patients, and 100 healthy subjects, GYPA, GYPB, GYPC, GYPE All four erythrocyte receptor genes were sequenced using a next-generation sequencer (NGS) and analyzed for nucleotide sequence polymorphisms. In addition, as reference strains of Plasmodium falciparum, 3D7, an African strain, and Dd2, an Indonesian strain, were also analyzed for the full nucleotide sequences of the three ligand genes, Eba-175, Eba-140, and Ebl-1. gone. Furthermore, the study is proceeding with the analysis of patient-derived malaria parasite gene polymorphisms.
